Gold Fibers as a platform for biosensing

A new form of high surface bioelectrode based on electrospun gold microfiber with-immobilized glucose oxidase was developed. The gold fibers were prepared by electroless deposition of gold nanoparticles on a poly(acrylonitrile)-HAuCl4 electrospun fiber. The material was characterized using electron microscopy, XRD and BET, as well as cyclic voltammetry and biochemical assay of the immobilized enzyme. The surface area of the gold microfibers was 2.5 m2/g. Glucose oxidase was covalently crosslinked to the gold surface using cystamine monolayer and glutardialdehyde, and portrayed characteristic catalytic currents for oxidizing glucose using a ferrocene methanol mediator. Limit of detection of glucose is 0.1 mM. The K m of the immobilized enzyme is 10 mM, in accordance with other reports of immobilized glucose oxidase. The microfiber electrode was reproducible and showed correlation between fiber weight, cathodic current and enzymatic loading.

Extracellular pH can indicate the variation in organelle function and cell state. It is important to measure extracellular pH (pHe) with a controllable distance. In this work, a potentiometric SECM dual-microelectrode was developed to monitor the pHe of MCF-7 cells under electrical stimulation. The distance between the dual-microelectrode and the cells was determined first with a gold microelectrode by recording the approaching curve, and the pH was determined using an open-circuit potential (OCP) technique with a polyaniline-modified Pt microelectrode. The pH microelectrode showed a response slope of 53.0 ¡À 0.4 mV/pH and good reversibility from pH 4 to pH 8, fast response within 10 s, and a potential drift of 1.13% for 3 h, and thus was employed to monitor the pHe of stimulated cells. The value of pHe decreased with the decrease in the distance to cells, likely due to the release of H+. With an increase in the stimulation potential or time, the pHe value decreased, as the cell membrane became more permeable, which was verified by fluorescence staining of calcein-AM/PI (propidium iodide). Based on these results, this method can be widely applied for determining the species released by biosystems at a controllable position.

A facile, highly stereo- and regioselective hydrometalation of alkynes generating alkenylmetal complex is disclosed for the first time from a reaction of alkyne, carboxylic acid, and a zerovalent group 10 transition metal complex M(PEt3)4 (M = Ni, Pd, Pt). A mechanistic study showed that the hydrometalation does not proceed via the reaction of alkyne with a hydridometal generated by the protonation of a carboxylic acid with Pt(PEt 3)4, but proceeds via a reaction of an alkyne coordinate metal complex with the acid. This finding clarifies the long proposed reaction mechanism that operates via the generation of an alkenylpalladium intermediate and subsequent transformation of this complex in a variety of reactions catalyzed by a combination of Bronsted acid and Pd(0) complex. This finding also leads to the disclosure of an unprecedented reduction of alkynes with formic acid that can selectively produce cis-, trans-alkenes and alkanes by slightly tuning the conditions.

Synthesis, structure and enantiomeric resolution of ferrocenylalkyl mercaptoazoles. Antitumor activity in vivo

Ferrocenylalkyl 2-mercaptobenzimidazoles 3 (a-e) and 2-mercaptobenzo[d]thiazole-2(3H)-thiones 5 (a-e) were prepared via the reaction of the alpha-(hydroxy)alkyl ferrocenes, FcCHR(OH) (1a-e; Fc = ferrocenyl; R = H, Me, Et, i-Pr, Ph), either with thiobenzimidazole in acetone at room temperature in the presence of TFA (catalytic amounts), in yields of 55-74%, or with thiobenzothiazole in methylendicloride in presence of aqueous HBF4 (equimolar amounts) at r.t.; in yields of 41-58%. The structures, electrochemical properties and enantiomeric resolution 3a-e and 5a-e (using HPLC on modified amylose as chiral selector) were investigated. In cyclic voltammetry all studied compounds exhibited a reversible one-electron oxidation-reduction wave owing to the ferrocene-ferricenium redox couple with a positive shift (0.56-0.80 V) compared with that of ferrocene (0.50 V). X-ray determinations of molecular structures of 3-ferrocenylmethylbenzo[d]thiazole-2(3H)-thione (5a) 3-ferrocenylethylbenzo[d]thiazole-2(3H)-thione (5b) and 3-ferrocenylphenylmethylbenzo[d]thiazole-2(3H)-thione (5d) were carried out. The toxicity and antitumor activity of N-(ferrocenylethyl)-2-thiobenzimidazole (3b) were evaluated in vivo. Maximum tolerated dose (MTD) value for the compound 3b was found to be equal to 800 mg kg-1. The effectiveness of compound under investigation against murine solid tumor system, carcinoma Ca755 (Ca755), was studied in a wide range of doses and significant antitumor effects were found. The index of tumor growth inhibition (TGI) on Ca755 equaled 87% in comparison with control.

The synthesis and characterization of a series of carbosilane-supported ferrocenyl phosphine ruthenium complexes of type SiMe4-n(Fe(eta5-C5H4SiMe2(CH2)3)((eta5-C5H4PR2)RuCl2(eta6-p-cymene)))n (p-cymene = 1-i-Pr-4-Me-C6H4; n = 2: 10a, R = Ph; 10b, R = cC6H11; 10c, R = 2-(5-Me)C4H2O); n = 4: 11a, R = Ph; 11b, R = cC6H11; 11c, R = 2-(5-Me)C4H2O)) is described. For comparative reasons, the non-immobilized ferrocenyl phosphine ruthenium complexes [FcPR2(RuCl2(eta6-p-cymene))] (Fc = Fe(eta5-C5H4)(eta5-C5H5); 9a, R = Ph; 9b, R = cC6H11; 9c, R = 2-(5-Me)C4H2O) were prepared. The molecular structure of 9c in the solid state is reported confirming the expected tetrahedral coordination sphere about the phosphorus atom and the “piano-stool” geometry about ruthenium. The ruthenium complexes 9-11 are catalytically active in the addition of benzoic acid to propargyl alcohol to form beta-oxopropyl benzoate. The obtained activities and productivities show that a good solubility of the catalyst is necessary for a successful catalytic reaction. Furthermore, the rate of the reaction can be influenced by using less basic and electron-withdrawing phosphine ligands.

Synthesis, characterization and antimicrobial evaluation of mono- and polynuclear ferrocenyl-derived amino and imino complexes

A series of ferrocenyl mono- and polynuclear complexes conjugated to imino and amino scaffolds was prepared. The imino complexes were prepared via a Schiff base condensation reaction between (E)-4-vinylferrocenylbenzaldehyde and various amines. The amino complexes were prepared by reductive amination reactions using the same precursors. The compounds were characterized using standard spectroscopic and analytical techniques including Nuclear Magnetic Resonance (NMR) spectroscopy, Infrared (IR) spectroscopy and mass spectrometry. The compounds were screened against Mycobacterium tuberculosis as well as the NF54 chloroquine-sensitive (CQS) strain of Plasmodium falciparum. Overall, the complexes showed moderate in vitro biological activity, with the ferrocenylimines exhibiting enhanced activity against M. tuberculosis compared to the corresponding amines. The ferrocenylimines also displayed moderate activity against P. falciparum, with the second-generation polyimine complex exhibiting the highest activity in vitro. The ferrocenylamines exhibit promising antiplasmodial activity, enhanced compared to the imines, with the silicon-containing derivative and the second-generation dendrimer showing good activity.
