Dispersion of multi-wall carbon nanotubes in glucose oxidase: Characterization and analytical applications for glucose biosensing

In this work we report for the first time the use of the enzyme glucose oxidase (GOx) to efficiently disperse multiwall carbon nanotubes (CNT) and to confer biorecognition properties to the dispersed nanotubes. The optimum dispersion was obtained by sonicating for 15 min 1.0 mg/mL CNT in 1.0 mg/mL GOx solution prepared in 50:50 ethanol/water. The dispersion was evaluated by Scanning Electron Microscopy (SEM), Infrared (FT-IR) and Ultraviolet-visible (UV-vis) Spectroscopy. The electrochemical characterization of glassy carbon electrodes (GCE) modified with the dispersion (by dropping) was performed by Electrochemical Impedance Spectroscopy (EIS), Cyclic Voltammetry (CV), and Amperometry. The amount of electroactive GOx deposited on GCE (GCE/CNT-GOx) was 1.02 ¡Á 10-10 mol cm-2 and the rate constant for the electron transfer between FAD center and the electrode was (2.9 ¡À 0.1) s-1 according to Laviron and (9.2 ¡À 1.3) s-1 considering the model proposed by Albery. The enzyme demonstrated to keep its biocatalytic activity even after dissolution in 50/50 v/v, ethanol-water solution and sonication for 15 min using either ferrocene methanol or oxygen as redox mediators. The sensitivity to glucose at 0.700 V obtained for seventeen electrodes prepared with 6 different dispersions was (3.2 ¡À 0.2) ¡Á 102 muA M-1, (r = 0.997), with an R.S.D. of 6.0%. The sensitivity remained highly constant after 30 days at room temperature (25 C) and 4 C, with average values of (3.21 ¡À 0.07) ¡Á 102 muA M-1, r = 0.9992 and (3.59 ¡À 0.08) ¡Á 102 muA M-1, r = 0.9990, respectively. The GCE/CNT-GOx can be used as platform to build supramolecular architectures for biosensing through the self-assembling of polyelectrolytes, opening the doors to new and exciting possibilities for the development of biosensors.

Label-Free Homogeneous Electrochemical Sensing Platform for Protein Kinase Assay Based on Carboxypeptidase Y-Assisted Peptide Cleavage and Vertically Ordered Mesoporous Silica Films

Presented herein is a simple, robust, and label-free homogeneous electrochemical sensing platform constructed for the detection of protein kinase activity and inhibition by integration of carboxypeptidase Y (CPY)-assisted peptide cleavage reaction and vertically ordered mesoporous silica films (MSFs). In this sensing platform, the substrate peptide composed of kinase-specific recognized sequence and multiple positively charged arginine (R) residues was ingeniously designed. In the presence of protein kinase, the substrate peptide was phosphorylated and then immediately resisted CPY cleavage. The phosphorylated peptide could be effectively adsorbed on the negatively charged surface of MSFs modified indium-tin oxide (ITO) electrode (MSFs/ITO) by noncovalent electrostatic attraction. The adsorbed peptide was subsequently used as a hamper to prevent the diffusion of electroactive probe (FcMeOH) to the electrode surface through the vertically aligned nanopores, resulting in a detectable reduction of electrochemical signal. As demonstrated for the feasibility and universality of the sensing platform, both protein kinase A (PKA) and casein kinase II (CK2) were selected as the models, and the detection limits were determined to be 0.083 and 0.095 UmL-1, respectively. This sensing platform had the merits of simplicity, easy manipulation, and improved phosphorylation and cleavage efficiency, which benefited from homogeneous solution reactions without sophisticated modification or immobilization procedures. In addition, given the key role of inhibition and protein kinase activity detection in cell lysates, this proposed sensing platform showed great potential in kinase-related bioanalysis and clinical biomedicine.

Influence of macroporous gold support and its functionalization on lactate oxidase-based biosensors response

A general bioanalytical platform for biosensor applications was developed based on three-dimensional ordered macroporous (3DOM) gold film modified electrodes using lactate oxidase (LOx) as a case study, within the framework of developing approaches of broad applicability. The electrode was electrochemically fabricated with an inverted opal template, making the surface area of the 3DOM gold electrode up to 18 times higher than that of bare flat gold electrodes. These new electrochemical transducers were characterized by using Field Emission Scanning Electron Microscopy (FE-SEM), Atomic Force Microscopy (AFM) and the X-ray diffraction (XRD). The biosensor was developed by immobilization of lactate oxidase (LOx), on a 3DOM gold electrode modified with a self-assembled monolayer of dithiobis-N-succinimidyl propionate (DTSP). The resulting lactate oxidase biosensor was characterized by electrochemical impedance spectroscopy (EIS). The 3DOM gold electrode not only provides a good biocompatible microenvironment but also promotes the increase of conductivity and stability. Thus, the developed lactate oxidase bioanalytical platforms showed higher mediated bioelectrocatalytic activity compared to others previously described based on polycrystalline gold transducers. The response to varying lactate concentrations has been obtained in the presence of hydroxymethylferrocene as redox mediator in solution. Under these conditions, the bioanalytical platform response for DTSP covalently bound enzyme was improved with respect to that obtained in absence of DTSP.

Diffusion of ferrocene methanol in super-cooled aqueous solutions using cylindrical microelectrodes

The diffusion of ferrocene methanol in super-cooled aqueous solutions containing sucrose has been studied, using disk and cylindrical microelectrodes, over a wide viscosity range. The solution viscosity and the reduced temperature T/Tg (Tg being the glass transition temperature) were varied by changing the sucrose concentration and the temperature of the system. The voltammetric limiting current obtained with a disk microelectrode and the i(t) response on a cylindrical microelectrode after a potential step were used to determine diffusion coefficients from 7 ¡Á 10-6 cm2 s-1 down to 2 ¡Á 10-11 cm2 s-1. The electrochemical procedure described in this work allows a simple and accurate measurement of the dynamics of electroactive solutes in glass-forming liquids.

Potentiometric sulfite biosensor based on entrapment of sulfite oxidase in a polypyrrole film on a platinum electrode modified with platinum nanoparticles

The surface?of a platinum electrode has been modified with platinum nanoparticles (PtNPs) and the enzyme sulfite oxidase (SOx), was entrapped on its surface in an ultrathin polypyrrole (PPy) film. The PtNPs, with a diameter of 30?40?nm, were deposited on the Pt electrode by cycling the electrode potential 20 times from -200 to 200?mV at a sweep rate of 50?mV.s-1. Morphological evidence of the successful incorporation of SOx and the presence of PtNPs were obtained by scanning electron microscopy. Also, the electrochemical behavior of the PtNPs/PPy-SOx film was examined by cyclic voltammetry, chronopotentiometry, electrochemical impedance spectroscopy and potentiometry. Under optimized conditions, the biosensor achieved a sensitivity of 57.5?mV.decade-1, a linear response that extends from 0.75 to 65?muM of sulfite, a detection limit of 12.4?nM, and a response time of 3?5?s. The biosensor was successfully applied to the determination of sulfite in wine and beer samples. [Figure not available: see fulltext.]

Copper-promoted azide-alkyne cycloadditions were attempted to synthesize a series of variedly functionalized 1H-1,2,3-triazole-linked isatin-ferrocene, ferrocenylmethoxy-isatin and isatin-ferrocenyl-chalcone conjugates. The synthesized scaffolds were assayed for their inhibitory activity against T. vaginalis as well as several common normal human flora bacteria. The observed inhibitory activities against T. vaginalis and undetectable inhibition of microflora bacteria suggest that these compounds may be specific against trichomonad protozoa and could serve as a new scaffold for synthesis of novel compounds against this important human pathogen.

Redox-flexible NADH oxidase biosensor: A platform for various dehydrogenase bioassays and biosensors

A generic amperometric bioassay based on the enzymatic oxidation catalysed by the stable NADH oxidase (NAox) from Thermus thermophilus has been developed for NADH measurements. The NAox uses O2 as its natural electron acceptor and produces H2O2 in a two-electron process. Electrochemical and spectrophotometric experiments showed that the NAox used in this work, presents a very good activity towards its substrate and, in contrary to previously mentioned NADH oxidases, does not require the addition of any exogenous flavin cofactor neither to promote nor to maintain its activity. In addition, the NAox used also works with artificial electron acceptors like ferrocene derivatives. O2 was successfully replaced by redox mediators such as hydroxymethyl ferrocene (FcCH2OH) for the regeneration of the active enzyme. Combining the NAox with the mediator and the horseradish peroxidase we developed an original, high sensitive “redox-flexible” NADH amperometric bioassay working in a large window of applied potentials in both oxidation and reduction modes. The biosensor has a continuous and complementary linearity range permitting to measure NADH concentrations starting from 5 ¡Á 10-6 M in reduction until 2 ¡Á 103 M in oxidation. This redox-flexibility allows choosing the applied potential in order to avoid electrochemical interferences. The association of the “redox-flexible” concept with NADH dependent enzymes opens a novel strategy for dehydrogenases based bioassays and biosensors. The great number of dehydrogenases available makes the concept applicable for numerous substrates to analyse. Moreover it allows the development of a wide range of biosensors on the basis of a generic platform. This gives several advantages over the previous manufacturing techniques and offers a general and flexible scheme for the fabrication of biosensors presenting high sensitivities, wide calibration ranges and less affected by electrochemical interferences.
